Modeling Clinical Development Strategies with Biomarkers at the FDA

A recently concluded cooperative R&D project at the FDA led to the development of a web-enabled software tool that will be useful for modeling the impact of predictive biomarkers on the complexity and cost of clinical development programs. Hear from the project's FDA Principal Investigator and from Adaptive Pharmacogenomic's PI on the project in a pair of webinars, August 24 and 25.

Pharmacogenetics of cytochrome P450 (CYP) in the elderly.

Pharmacogenetics of cytochrome P450 (CYP) in the elderly.

Ageing Res Rev. 2010 Jun 20;

Authors: Seripa D, Pilotto A, Panza F, Matera MG, Pilotto A

The genetics of cytochrome P450 (CYP) is a very active area of multidisciplinary research, overlapping the interest of medicine, biology and pharmacology, being the CYP enzyme system responsible for the metabolism of more than 80% of the commercially available drugs.

Drug interactions with HMG-CoA reductase inhibitors (statins): the importance of CYP enzymes, transporters and pharmacogenetics.

Curr Opin Investig Drugs. 2010 Mar;11(3):323-32

Authors: Neuvonen PJ

HMG-CoA reductase inhibitors (statins) can cause skeletal muscle toxicity; the risk of toxicity is elevated by drug interactions and pharmacogenetic factors that increase the concentration of statins in the plasma. Statins are substrates for several membrane transporters that may mediate drug interactions.

Clopidogrel resistance.

Am J Ther. 2010 Mar-Apr;17(2):210-5

Authors: Sugunaraj JP, Palaniswamy C, Selvaraj DR, Chaitanya Arudra SK, Sukhija R

Clopidogrel resistance is an emerging clinical scenario, as antiplatelet therapy has become the cornerstone of modern cardiovascular treatment.

Ethical and social issues in pharmacogenomics testing.

Curr Pharm Des. 2010;16(2):245-52

Authors: Vijverberg SJ, Pieters T, Cornel MC

In genomics research, pathways that lead to disease and the role of drugs in these pathways are being unravelled at a high rate. In this paper ethical and social challenges related to pharmacogenomics research are discussed as well as clinical applications.

Pharmacogenetics of oral anticoagulant therapy.

Curr Pharm Des. 2010;16(2):187-203

Authors: Schalekamp T, de Boer A

The identification of the genes encoding CYP2C9, the principal metabolizing enzyme of the coumarins, and VKORC1, the molecular target for coumarins, has strongly stimulated the research on pharmacogenetics of vitamin K antagonists, also designated as coumarins.

Pharmacogenetics of thiopurines in inflammatory bowel disease.

Curr Pharm Des. 2010;16(2):145-54

Authors: Derijks LJ, Wong DR

Thiopurines are widely used in the treatment of inflammatory bowel disease (IBD). However, in clinical practice azathioprine (AZA) or 6-mercaptopurine (6-MP) are not effective in one-third of patients and up to one-fifth of patients discontinue thiopurine therapy due to adverse reactions.

Pharmacogenetics of phase I and phase II drug metabolism.

Curr Pharm Des. 2010;16(2):204-19

Authors: Crettol S, Petrovic N, Murray M

Genetic variation in the receptors and other intracellular targets that mediate the pharmacodynamic effects of drugs can affect therapeutic outcomes.

Pharmacogenetics in psychiatry--a useful clinical tool or wishful thinking for the future?

Curr Pharm Des. 2010;16(2):136-44

Authors: Kirchheiner J, Seeringer A, Viviani R

More than fifty years of pharmacogenetic research have produced many examples of the impact of inherited variability in the response to psychotropic drugs. These successes, however, have as yet failed to translate into broadly applicable strategies for the improvement of individual drug treatment in psychiatry.

Pharmacogenetics of drug transporters.

Curr Pharm Des. 2010;16(2):220-30

Authors: Franke RM, Gardner ER, Sparreboom A

During the last decade, a greater focus has been given to impact of genetic variation in membrane transporters on the pharmacokinetics and toxicity of numerous therapeutic drugs.
